Menadione, commonly known as vitamin K3, is a synthetic form of vitamin K that plays a critical role in blood coagulation and bone metabolism. Unlike naturally occurring vitamin K1 (phylloquinone) and K2 (menaquinone), menadione is produced synthetically and primarily used in animal feed, though its biological activity and potential therapeutic properties continue to attract scientific interest.
Primary Applications of Menadione in Animal Feed
The most widespread use of menadione is as a vitamin K source in livestock and poultry nutrition. Menadione sodium bisulfite complex (MSBC) is the standard form added to commercial animal feeds, providing essential vitamin K activity that supports proper blood clotting and bone health in animals.
In 2021, the Association of American Feed Control Officials (AAFCO) convened an expert panel that confirmed MSBC as a safe and suitable source of vitamin K for all animal species in the United States. This recommendation was supported by confidential industry safety data, independent literature reviews, and the European Food Safety Authority’s scientific opinion.
MSBC and the related compound menadione nicotinamide bisulfite (MNB) are authorized for use in animal feed across the European Union, Canada, and the United States. Their inclusion helps prevent hemorrhagic conditions caused by vitamin K deficiency, which can arise from inadequate diet, antibiotic use disrupting gut microbiota, or ingestion of vitamin K antagonists.
Menadione’s Role in Vitamin K Metabolism
Research has shown that menadione acts as a metabolic intermediate in the vitamin K pathway. Studies in rats demonstrated that orally administered phylloquinone (vitamin K1) is converted to menadione in the intestine, which then circulates as a precursor and is converted to menaquinone-4 (vitamin K2) in various tissues.
This conversion pathway highlights menadione’s biological importance, even though the compound itself is not used as a dietary supplement for humans. The liver can convert menadione into active vitamin K2, which participates in gamma-carboxylation reactions essential for producing functional clotting factors and bone proteins like osteocalcin.
Why Menadione Is Not Used in Human Supplements
Despite its biological activity, menadione is not legally sold as a human dietary supplement in most countries. Research dating back to the 1980s and 1990s demonstrated that menadione can damage red blood cells and cause liver toxicity in humans at certain doses.
For this reason, only vitamins K1 and K2 are available as human supplements and prescription medications. The U.S. National Academy of Sciences and the European Food Safety Authority have not established an upper intake limit for natural forms of vitamin K due to their low toxicity potential.
Emerging Research on Anti-Cancer Properties
Although toxic to humans at supplemental doses, menadione has shown promising anti-cancer and anti-inflammatory properties in laboratory studies. Test-tube research found that vitamin K3 can kill human breast, colorectal, and kidney cancer cells by activating specific proteins and increasing the production of reactive oxygen species.
These findings are still in early stages, and no clinical applications for humans have been approved. However, the research underscores the complex pharmacological profile of this synthetic vitamin K form.
